Immunohistochemistry.Paraffin-embedded samples were sectioned at 5 μm. After being deparaffinized and rehydrated, the samples were autoclaved in 1 mM HCl at 121°C for 5 min and subsequently washed with PBS. The samples were digested with 50 μg/ml PK in PBS at 37°C for 30 min, treated with 3 M guanidine thiocyanate at RT for 10 min, and then washed with PBS. After blocking with 5% FBS in PBS at RT for 1 h, the samples were incubated with 6D11 anti-PrP Ab at RT for 2 h and washed with PBS. The samples were then treated with ImmPRESS reagent anti-mouse IgG (MP-7402; Vector Laboratories, Burlingame, CA) at RT for 1 h. After washing with PBS, the samples were incubated with ImmPACT DAB peroxidase substrate (SK-4105; Vector Laboratories) for 180 s for staining.
RD cells were preseeded in 96-well plates with 1 × 104 cells per well. Antisera or mAb 2G8 was serially diluted twofold and incubated with an equal volume of CVA10 (100 TCID50 per well) at 37°C for 1 hour. The mixtures were then added to cells and incubated at 37°C for 5 to 7 days. The cytopathic effect (CPE) was observed with microscopy, and a neutralizing titer was defined as the highest dilution giving >50% neutralization of the well. A neutralizing titer of antisera with values of ≥1:8 was considered as a threshold for positivity.
The as-prepared BP was resuspended into a Teflon autoclave with 20 ml of water and treated at 200°C for 6 hours. After cooling down to room temperature, the DP was fabricated and washed in 50% alcohol solution, followed by dispersion into 1 ml of 50% alcohol solution for storage.
In addition, PG-Triazole-P was prepared in DMF (30.27 ml) from PGA (0.150 g, 1.51 × 10−3 mol azido group) and phenylacetylene (0.17 g, 1.66 × 10−3 mol). The polymer was obtained as a pale-green solid. Yield: 74%. 1H NMR (300 MHz, CDCl3, δ (p.p.m.)): 8.197–7.479 (br, 3H, triazole, aromatic proton), 7.358–6.937 (br, 3H, aromatic proton), 4.644–3.056 (br, 5H, OCH, OCH2, -CH2-triazole) (Supplementary Figure S2c). In the analysis of the abundance of barn swallows, we used generalized linear models, assuming a Poisson distribution of count data. As predictor variables, we included log10-transformed radiation and all potentially confounding variables listed above. In addition, we included temperature, cloud cover, wind, time of day and time of day squared, the latter to account for the fact that bird activity has a peak during early morning and a second, milder peak in the afternoon. We also included radiation level squared to account for non-linear relationships between species richness and abundance, respectively, and radiation. These analyses were all implemented in the statistical software JMP (SAS Institute Inc., 2012). In the analyses of the proportion of swallows being juveniles, we relied on general linear models with binomially distributed data and a logit link function.
